It was founded in 1607 and was the brainchild of Archbishop Federico Borromeo, who was passionate about science and art from a young age. Today it is one of the jewels of Milan and Italy. Its Baroque building is located in the center of the city and was built by Lelio Buzzi and Francesco Maria Richini; it houses one of the richest collections of Greek, Latin, and Oriental manuscripts, as well as Leonardo Da Vinci's Atlantic Codex.

It is one of the largest libraries in Italy, as well as one of the most beautiful, and is also located at the Brera Academy. It was founded in 1770 by Maria Theresa of Austria because she considered the Ambrosiana library rich in manuscripts but poor in printed books. It has about 1.5 million volumes that find space among the shelves in the Maria Theresa Room, designed by Giuseppe Piermarini in walnut wood. The Bohemian crystal chandeliers were reconstructed from the remains of those in the Caryatid Hall of the Royal Palace, which were destroyed during World War II.
